We were picked up in the French Quarter. If you let them know where you are staying, they will make arrangements for you.
Regarding schedules and traffic on a Friday. Would it be better to do the noon or 2:45 tour, with hotel pick up?

The noon pick up would be much better. It takes at least an hour to get to the boats before you can enjoy your tour, thus being back early evening to enjoy your evening.
